You can start your bot out on den of evil and blood raven, and work your way up from there.
Unfortunatly, you will have to personally go to the Tree of Infuniss and get the scroll to get to trist. THe bot only runs what you have available, and you'd have to personally do the quests.
Can't say about a downside on ladder, other than the extra runewords you can made
